You would predict a stronger, longer version of the natural signal. Endocrinology had other plans.
Because this axis reads rhythm rather than volume, a signal that will not go away is not a louder message. It is a jammed frequency.

Glycine out, D-tryptophan in. The D form is the mirror image of what peptidases are built to cut, so the molecule survives where native GnRH gets chopped in half. Everything else in the sequence is untouched.
An agonist agonises first. LH and FSH rise early, then sustained occupancy downregulates the receptor and output drops below where it started. That initial rise is the mechanism working, not a fault in it.
Rung once an hour, the pituitary keeps answering. Held down, it stops. Belchetz and colleagues showed that in 1978 and credited the pattern of delivery rather than the amounts.

The 1976 Journal of Medicinal Chemistry paper reporting D-Trp6 LH-RH at thirteen times the activity of LH-RH, and the 1978 Science paper establishing that pattern beats amount.
Any of that transferring to a research vial. The clinical file belongs to specific depot formulations with defined release profiles, not to lyophilised powder sitting on a bench. There's no ambiguity on the sport side either, since WADA names triptorelin outright in section S2.2.1 of the Prohibited List.
The defining experiment for this molecular class is Belchetz, Plant, Nakai, Keogh and Knobil, Science 1978, PMID 100883, conducted in rhesus monkeys with hypothalamic lesions that eliminated endogenous GnRH input and left exogenous delivery as the only variable. Intermittent administration at one pulse per hour reestablished pituitary gonadotropin secretion, whereas continuous infusion failed to restore sustained secretion, and the authors attribute the difference to the pattern of GnRH delivery rather than to the amounts administered, noting that continuous administration can result in desensitisation or down regulation. Triptorelin's clinical utility follows directly from that result. Substituting D-tryptophan at position six confers resistance to enzymatic cleavage, and depot formulation extends exposure further, so the receptor is held in the continuous condition rather than the pulsatile one. The consequence is suppression of the hypothalamic pituitary gonadal axis after an initial agonist surge, which is the basis of its use in advanced prostate cancer and central precocious puberty. One receptor, opposite endocrine outcomes, decided by the temporal pattern of occupancy.

Schally's lab had already spent a decade proving what GnRH was. The obvious next move was to build a better one, and in 1976 better meant stronger.
The trick they reached for is one of the oldest in peptide chemistry. Your enzymes evolved to recognise L amino acids, because that's what proteins are made of.
Hand them a D amino acid, the mirror image, and the scissors don't close properly. Put that D residue exactly where the peptide normally gets cut, which for GnRH is position six, and the molecule survives a great deal longer in circulation.

Position six. Gonadorelin has glycine there and is identical to your own GnRH. Triptorelin has D-tryptophan and lasts far longer.
