Triptorelin is a synthetic decapeptide GnRH agonist with the sequence pGlu-His-Trp-Ser-Tyr-D-Trp-Leu-Arg-Pro-Gly-NH2, differing from gonadorelin only by substitution of D-tryptophan for glycine at position six, with free base formula C64H82N18O13, molecular weight 1311.4 and CAS number 57773-63-4. Continuous receptor occupancy from depot formulations desensitises pituitary GnRH receptors and suppresses gonadotropin secretion, and triptorelin is authorised in Canada as a prescription product under DINs including 02389282 and 02240000.
One substitution. Position six. A D-tryptophan, the mirror-image form, dropped into GnRH's ten residues.
Result: a molecule about thirteen times more potent in the classic rat assays, and far more resistant to being cleared.
You would predict a stronger, longer version of the natural signal. Endocrinology had other plans.
Because this axis reads rhythm rather than volume, a signal that will not go away is not a louder message. It is a jammed frequency.

Triptorelin GnRH backstory
How it got here.
Schally's lab had already spent a decade proving what GnRH was. The obvious next move was to build a better one, and in 1976 better meant stronger.
The trick they reached for is one of the oldest in peptide chemistry. Your enzymes evolved to recognise L amino acids, because that's what proteins are made of. Hand them a D amino acid, the mirror image, and the scissors don't close properly. Put that D residue exactly where the peptide normally gets cut, which for GnRH is position six, and the molecule survives a great deal longer in circulation.
Coy, Vilchez-Martinez, Coy and Schally reported the result in 1976. Thirteen times more active than LH-RH itself in immature male rats. A superagonist, on purpose.
The part nobody planned for is what the field did with it. Within a few years urologists and paediatric endocrinologists were using it not to turn the axis up but to shut it off, because sustained stimulation of that receptor produces the opposite of pulsed stimulation. The most potent fertility signal anybody had synthesised became a fertility suppressant. Same molecule, different clock.

How can an agonist end up suppressing hormones?
Because the receptor responds to timing and not just to occupancy. Pulsed stimulation keeps gonadotropin secretion running, while continuous stimulation desensitises and downregulates the receptor until output falls below where it started. Belchetz and colleagues demonstrated exactly that in Science in 1978. Depot formulations of triptorelin are engineered to hold the continuous condition, so a stimulator behaves like a suppressor.
Is triptorelin approved in Canada?
Yes. Health Canada shows marketed DINs 02389282 for Decapeptyl from Ferring and 02240000, 02243856 and 02412322 for Trelstar from Knight, all of them prescription. FDA approved Trelstar under NDA 020715 in 2000 and Triptodur under NDA 208956 in 2017. Those approvals belong to specific depot products, not to the research vial on this page.
What's the difference between triptorelin and gonadorelin?
Position six. Gonadorelin has glycine there and is identical to your own GnRH. Triptorelin has D-tryptophan and lasts far longer. That difference in duration flips the outcome, because a short signal reads as a pulse and a long one reads as continuous. One of them turns the reproductive axis on, the other one eventually turns it off.
Is triptorelin banned in sport?
Yes, named explicitly in section S2.2.1 of the WADA Prohibited List, sitting right alongside gonadorelin. That category covers agents affecting the hypothalamic pituitary gonadal axis, and triptorelin is one of the compounds spelled out by name rather than swept in under a catch all clause.
