Gonadorelin is a synthetic decapeptide identical to endogenous gonadotropin releasing hormone, sequence pGlu-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH2, with molecular formula C55H75N17O13, molecular weight 1182.3 and CAS number 33515-09-2. No human gonadorelin product is currently marketed in Canada or the United States, as the FDA registrations Factrel and Lutrepulse are discontinued and all Canadian human DINs are cancelled or dormant, leaving only veterinary preparations on the market.
This is not an analogue of anything. Gonadorelin is GnRH, the genuine ten-residue hormone, sequence-identical to what your hypothalamus releases.
Characterising it won the 1977 Nobel Prize. This molecule is a chapter heading in endocrinology.
It sits one step below kisspeptin in the reproductive chain of command: it carries the order from brain to pituitary, in pulses, roughly every ninety minutes.
And here is the strangest fact on this page: check the Canadian drug registry and every human gonadorelin product is cancelled.
The marketed DINs that remain are veterinary. In this country, the molecule that won a Nobel for human endocrinology is officially cattle medicine.

GnRHR is a GPCR sitting on pituitary gonadotroph cells. Gq, then phospholipase C, then calcium, then the cell releases its stored LH and FSH. Short chain, well mapped, no mystery left in it.
Belchetz and colleagues, Science 1978, in hypothalamically lesioned rhesus monkeys. Hourly pulses reestablished gonadotropin secretion. Continuous infusion failed to sustain it. The abstract credits the pattern of delivery rather than the amounts.
Gonadorelin clears fast because your body built it to. That's a feature for pulsatile work and a nuisance for everything else, and it's precisely why the long acting analogues got invented.
The pulse is the message. This axis reads rhythm, not volume.
This axis runs on the most famous rhythm lesson in endocrinology, and it is worth learning properly.
Pulsed GnRH, arriving every hour or two, keeps the pituitary releasing its hormones normally. Continuous GnRH exposure does the opposite: the receptors downregulate and the axis shuts off.
Same molecule. Same receptor. Opposite outcomes, decided entirely by timing.
That paradox is not a footnote, it is clinically exploited on purpose by the long-acting agonists, which suppress the axis by refusing to pulse.
Here's the honest part, and it writes itself.
A signal whose meaning inverts with its rhythm is a signal that punishes crude use. The pulse is not a detail of the mechanism. It is the mechanism.
The research so far for Gonadorelin Peptide
Where the evidence is thin, we say so.
The 1977 Nobel Prize in Physiology or Medicine for the discovery of the hypothalamic releasing hormones. A confirmed decapeptide sequence identical to endogenous GnRH, with PubChem and the FDA Factrel label agreeing on C55H75N17O13 and 1182.3. Decades of literature on the GnRH stimulation test and on pulsatile pump therapy for hypothalamic amenorrhoea.
A marketed human product. Factrel and Lutrepulse are discontinued at FDA, every Canadian human DIN is cancelled or dormant, and the only gonadorelin DINs Health Canada shows as marketed today are veterinary. There's no exemption hiding in that gap either, since WADA names gonadorelin in S2.2.1 no matter what's on a shelf.
Gonadorelin occupies an unusual regulatory position, in that the endogenous ligand is commercially extinct in North American human medicine while its synthetic analogues remain widely marketed. FDA lists Factrel, NDA 018123, and Lutrepulse, NDA 019687, as discontinued. Health Canada shows every human gonadorelin DIN cancelled or dormant, including 00368385, 02042096 and 02041820 for Factrel, 02046202 and 02046210 for Lutrepulse, the latter cancelled in December 2024, and Relisorm cancelled in 2019. The only gonadorelin DINs currently marketed in Canada are veterinary preparations used in bovine reproductive management. The displacement is mechanistic as much as commercial. Native GnRH has a short circulating half life and therefore reproduces physiological pulsatility, which requires pump hardware to exploit clinically, whereas substituted analogues achieve sustained receptor occupancy from a depot injection and produce suppression on a schedule a clinic can actually run. Gonadorelin remains explicitly named in section S2.2.1 of the WADA Prohibited List regardless of its marketing status.

How it got here.
Two labs spent the sixties chasing the same ghost. Everybody was convinced the hypothalamus talked to the pituitary chemically, and nobody could catch the messenger, because catching it meant working through a mountain of tissue to end up with a few milligrams of something that acts at concentrations you can barely measure.
Roger Guillemin and Andrew Schally raced each other to it, and not warmly. When the structure finally landed in 1971 it was ten residues long, which has to have felt anticlimactic for something that ate a decade of two careers.
The 1977 Nobel Prize in Physiology or Medicine went to the pair of them, sharing it with Rosalyn Yalow, whose radioimmunoassay is the technique that made hunting these things possible at all. The punchline shows up fifty years later.
The molecule they bled for has no human product on the market anywhere in North America, and the brand name Factrel is now printed on a bottle in a cattle vet's fridge.

One residue, and a completely opposite job. Triptorelin is the same decapeptide with D-tryptophan swapped in at position six, which makes it resistant to breakdown and long acting. Native gonadorelin clears fast, so it reads as a pulse. Sustained occupancy desensitises the receptor instead of stimulating it, so one of them starts the axis and the other shuts it down.
Yes, named explicitly in section S2.2.1. Worth knowing, because people assume a discontinued product quietly falls off the radar. It doesn't. The Prohibited List tracks substances and not marketing status, and gonadorelin's entry sits right next to triptorelin's. If you're subject to testing, treat it that way regardless of what a pharmacy stocks.

The human uses were a diagnostic challenge test and pulsatile pump therapy for hypothalamic amenorrhoea, and the pump route needed hardware most clinics had no appetite to run.
